报告摘要:Microalgae have significant potential as light-driven cell factories for the synthesis of a wide range of bioproducts. These include bioactive metabolites, high-value pigments, speciality enzymes, therapeutic proteins, biopolymers and RNA-based anti-viral agents. The genetic engineering of algae is most advanced for the model species Chlamydomonas reinhardtii, with well-established methods for manipulating both the nuclear and chloroplast genomes. The latter is particularly attractive as a site for insertion of foreign genes for a number of reasons: i) genes can be targeted to precise loci; ii) stable, high level expression can be achieved; iii) the small, low complexity genome is particularly suited to synthetic biology (synbio) approaches that allow rapid, standardised and predictive strain engineering; iv) the organelle is the site of numerous biosynthetic pathways and therefore represents the obvious‘chassis’on which to bolt new metabolic pathways that divert the carbon fixed by photosynthesis into novel compounds. I will present our recent progress on the development of synbio tools for genetic engineering of the chloroplast of C. reinhardtii, including a simple‘one-pot’method for DNA assembly and selection methods that avoid the use of antibiotic-based markers. Our tools also include design strategies for biocontainment of transgenes, and their high-level and inducible expression, together with a method for low-cost algal cultivation in non-sterilised media. I will give examples of applications in the field of therapeutic proteins including the production of anti-bacterial enzymes that target major human pathogens, and bio-encapsulated vaccines for oral delivery in the aquaculture industry.
